LIC Duet™ Adaptor cloning is a powerful method specifically designed for simultaneous, directional cloning of two open reading frames into Novagen Ek/LIC bacterial expression vectors. All LIC Duet Adaptors contain the T7lac promoter, a strong ribosome binding site (rbs), and an ATG translation initiation codon to drive expression of the second ORF. Four of the five adaptors allow for expression of the second ORF fused with an amino-terminal tag plus enterokinase cleavage site. The LIC Duet Minimal Adaptor encodes the minimum amino-terminal fusion sequence (five amino acids) with no protease cleavage site.
The adaptor sequences are provided in GenBank format, with significant features indicated.
