M13mp18 classic single stranded phage vector
When used as a T-Vector, there is an additional T in the Sma I site of the M13mp18 Vector such that the vector sequence reads CCCT before the cloned insert on both strands. The vector sequences are the same as Genbank accession #X02513, except that a single T residue was added at position 898 to restore the reading frame of gene V as in wild-type M13 (F. Pfeiffer, MPI, Martinsreid). The vector is numbered beginning at the first A of a Hinc II site that was modified during construction (Yanisch-Perron, C., Vieira, J. and Messing, J. (1985) Gene 33, 103-119). All coding sequences are transcribed clockwise. Only the + strand product (shown) of single-stranded replication is packaged into virions; therefore, sequencing of single-stranded DNA should be performed with the U-19mer primer.
