Protein-protein interactions are a key element in understanding the complex cellular network. To study these types of interactions, methods like yeast two-hybrid screens have been used. Though powerful, they also yield a substantial amount of false results. Another common method is tandem affinity purification (TAP) that requires two successive purification steps.

The One•StrEP™ Protein Interaction Kit improves on these methods by using the extremely fast and efficient One•StrEP•Tag/Strep•Tactin technology. Unlike the other systems, the One•StrEP™ Protein Interaction Kit requires only one step to isolate intact protein complexes with minimal background. After loading the cell extract to a Strep•Tactin column and washing away non-binding proteins, the recombinant protein (bait) and all bound proteins are gently eluted intact by the addition of 2 mM biotin. The proteins are then resolved on an SDS-gel and the bound proteins can be identified by mass spectrometry or by Western blot using specific antibodies.
