pCDF and pRSF Vectors
The pCDF and pRSF vectors are designed specifically to minimize the number of extraneous amino acids encoded when expressing amino-terminal His•Tag® fusion proteins. These vectors are available as 1) uncut plasmids with an extensive multiple cloning site (MCS) for restriction enzyme cloning methods, or 2) as linearized vectors prepared for ligation-independent cloning (LIC), or 3) as uncut Duet™ plasmids for restriction enzyme cloning of two independent ORFs.

  • pCDF vectors carry the CloDF13 replicon (1, 2), lacI gene and streptomycin/spectinomycin resistance gene (aadA).
  • pRSF vectors carry the RSF1030 replicon (3, 4), lacI gene and kanamycin resistance gene.

The vector sequences are provided in GenBank format, with significant features indicated. To convert the sequence into other formats, first select the sequence and copy (Command-C on Mac or Ctrl-C on Windows). Next, click here to open the NIH ReadSeq web page in a new window. Paste the sequence into the input field (Command-V on Mac or Ctrl-V on PC), choose your favorite format from the dropdown menu, and hit the "submit" button.
