Anti-Insulin Receptor Substrate-1, Rat (Mouse) Cat. No.: GR45 Host: Mouse Isotype: IgG Immunogen: A synthetic peptide (YYASINFQKQPEDRQ) corresponding to amino acid residues 1221 - 1235 of rat insulin receptor substrate-1 (IRS-1). Positive Control: HepG2 or 3T3-L1 cells or liver or muscle tissue. Negative Control: Normal Mouse Serum (Cat. No. NS03L) Comments: Recognizes a protein of 160-185 kDa depending on the cell type studied. IRS-1 is a major endogenous substrate for the insulin receptor and the IGF-IR tyrosine kinase.

Detects a band at ~185 kDa corresponding to IRS-1. Upon insulin stimulation, IRS proteins are phosphorylated by insulin receptor tyrosine kinase. Decreased levels of IRS proteins are associated with insulin resistance in humans.
CHO/IR cells, unstimulated (-) and stimulated with insulin (+) were lysed and either immunoprecipitated with anti-IRS-1 or IRS-2 (as indicated) and then blotted with anti-IRS-1 (left four columns). Antibody is highly specific for IRS-1 and does not detect protein precipitated by IRS-2. Immunoprecipitation removes most all IRS-1 from the cell lysate (right 2 columns).
